I have not read the books so I don't know. My bet is on his mum, Cersei. She hates Margaery and the fact she was going to be queen. She gets to frame Tyrion who she hates. She is the cruel enough, and remember she had the poison in the Blackwater episode she was going to poison Joffrey with to keep him from having to suffer had the hands of Stannis.

No way it's Cersei Lannister - there were a couple of scenes where she states her love for her son despite that he's become a monster. Besides, she could have just had Margarey Tyrell killed. Ok- definitely not Tyrion. Not Cercei. The cup of wine was grabbed from in front of the grandma, but you know what? I'm not sure it was the wine at all- I think it was the pie. Wasn't he the only one eating, whereas everyone was imbibing? I actually think it was probably Margaery herself. The pie seems possible as well, though if you're planning to poison someone, the pie would likely be more difficult to pull off. How do you know Joffrey will eat first? The most obvious suspect is the prince dude who had that exchange with Cersei and papa Lannister at the wedding. Dude basically said he was there to kill Lannisters, but that probably makes him too obvious.So Russell...what do you love about music? To begin with, everything.

The only ones we see on camera holding the goblet are Joffrey, Tyrion, Sansa, and Margaery (although she sets it down near Grandma Tyrell). Also Ser Dantos the fool seemed to know a bit too much about what was going on as he wisped Sansa away.

This could support Bruiserstone's theory. When Cersei and Tywin are talking to that prince dude, he keeps deliberately referring to her as the "former" queen regent. And then when Cersei tells the worker guy that the leftover food is for the hounds, he says "but the queen said," and she retorts, "no, the queen is telling you." Obviously she was going to look to reassert herself, so she has some motivation here. But I agree with JL that it just seems more likely she'd poison her own kid instead of just going after Margaery. Maybe the poison was in a mug intended for Margaery or the grandma and just accidentally ended up with Joffrey? That seems unlikely, though, with the way Sansa was whisked out of there.
